苦瓜藤中三萜类化合物的分离

【摘要】:苦瓜中的三萜苷元和三萜皂苷是苦瓜的有效成分,国内外研究表明,苦瓜中的三萜类物质具有改善胰岛素抵抗、降血糖、提高免疫力等作用。本论文以苦瓜藤为研究对象,以分离纯化三萜类化合物为研究目标,最终建立一种快速大量制备三萜类化合物的方法。本文首先利用正/反相硅胶柱层析色谱、MCI柱层析色谱、制备液相色谱等方法从苦瓜藤75%乙醇-水提取物中分离纯化并鉴定出6种三萜类化合物,分别是3,7,25-Trihydroxycucurbita-5,23-dien-19-al(化合物1)、momordicines I(化合物2)、葫芦烷-5,23-二烯-3β,7,25-三羟基-9-醛基-7-O-β-D-吡喃葡萄糖甙(3)、Momordicine IV(4)、Momordicine II(5)、3,7-二羟基-23(R)-乙氧基-19-醛基-5,24-二烯醇(6)。然后通过静态吸附和解吸附试验,对7种大孔树脂富集化合物1和化合物2的能力进行比较,筛选出能有效富集两种化合物的树脂HPD200A,并进一步研究了HPD200A树脂富集两种化合物的机理,结果表明化合物1和化合物2在HPD200A树脂上的吸附机理符合拟一阶吸附动力学模型。同时,韦伯-莫里斯模型表明化合物1和化合物2在HPD200A树脂上的吸附速率可能是由膜扩散和粒子内部扩散等方式决定。其次,通过研究苦瓜藤提取液中化合物1和化合物2在HPD200A树脂上的动态吸附和解吸附特性,确定了最优上样条件为:上样液中化合物浓度分别为0.068mg/ml,0.091mg/ml;上样液流速为3.5ml/min;上样液体积为树脂体积的3倍。最优洗脱条件依次为:蒸馏水(3倍树脂体积),60%乙醇-水溶液(4倍树脂体积),85%乙醇-水溶液(8倍树脂体积)。在此洗脱条件下,经过HPD200A树脂柱层析,制备的粗品杂质大量减少,化合物1和化合物2的含量分别从1.19%和1.60%增加到19.44%和21.91%。粗品经过制备液相纯化,得到5.3mg化合物1和7.1mg化合物2,经液相检测纯度均达到85%以上。
[Abstract]:Triterpenoid glycosides and triterpenoid saponins in balsam pear are effective components of balsam pear. Studies at home and abroad have shown that triterpenoids in balsam pear can improve insulin resistance, reduce blood sugar and enhance immunity. In this paper, balsam pear vine was taken as the research object, and the separation and purification of triterpenoids was taken as the research objective, and finally a rapid and large-scale preparation was established. Firstly, six triterpenoids, 3,7,25-Trihydroxycurbita-5,23-dien-19-al (compound 1), momordicines I (compound 2), were isolated and purified from 75% ethanol-water extract of Momordica charantia L. Cucurbitane-5,23-diene-3 beta,7,25-trihydroxy-9-aldehyde-7-O-beta-D-pyranoside(3),Momordicine IV(4),Momordicine II(5),3,7-dihydroxy-23(R)-ethoxy-19-aldehyde-5,24-dienol(6). The ability of seven macroporous resins to enrich compounds 1 and 2 was compared by static adsorption and desorption tests. The adsorption mechanism of compound 1 and compound 2 on HPD200A resin conformed to pseudo-first-order adsorption kinetics model. Meanwhile, Weber-Morris model showed that compound 1 and compound 2 were on HPD200A resin. The adsorption rate may be determined by membrane diffusion and particle internal diffusion. Secondly, the dynamic adsorption and desorption characteristics of compounds 1 and 2 in the extract of Momordica charantia L. on HPD200A resin were studied, and the optimum conditions were determined as follows: the concentration of compounds in the sample solution was 0.068 mg/ml, 0.091 mg/ml, and the flow rate of the sample solution was 3.5 ml/ml. The optimum elution conditions were: distilled water (3 times resin volume), 60% ethanol-water solution (4 times resin volume) and 85% ethanol-water solution (8 times resin volume). Under these elution conditions, the impurities of the crude products were greatly reduced and the contents of compounds 1 and 2 were respectively reduced by HPD200A resin column chromatography. From 1.19% and 1.60% to 19.44% and 21.91%. The crude product was purified by preparation liquid phase and 5.3 Mg compound 1 and 7.1 Mg compound 2 were obtained. The purity of the crude product was over 85% by liquid phase detection.
