卷烟烟气暴露对大鼠学习记忆功能的影响

【摘要】:研究卷烟烟气暴露对大鼠学习记忆功能的影响,并探讨脑源性神经营养因子(BDNF)的作用。将雄性SD大鼠随机分为对照组、低剂量组(染毒10min)、中剂量组(染毒20min)及高剂量组(染毒40min),置于气体染毒箱内行被动吸烟,分别于烟气暴露后的30d、90d、180d进行Morris水迷官实验,检测大鼠学习记忆功能,观察指标包括:动物体重,外周血血细胞参数,血清中同型半胱氨酸(Hcy)含量,HE染色观察海马区病理,免疫组化方法观察海马BDNF表达变化。 大鼠卷烟烟气暴露后,所检测各个指标与正常对照组相比较,结果如下: (1)大鼠体重增长明显下降; (2)白细胞数先增加后减少,红细胞数则呈先减少后增加的趋势; (3)染毒30d,烟气暴露组潜伏期均显著低于对照组(P0.05);染毒90d,中剂量烟气暴露组潜伏期显著低于对照组(P0.05);染毒180d,烟气暴露组潜伏期显著高于对照组(P0.05)。同组间随时间的增加,对照组表现为学习记忆功能逐渐增强,低剂量组为先降低后增强,中剂量组为增强,高剂量组无明显变化。 (4)HE染色结果表明卷烟烟气暴露30d、90d、180d大鼠大脑海马区无明显变化; (5)染毒30d至90d,低剂量组血清中Hcy含量增加,中剂量组和高剂量组含量下降;染毒180d,染毒组血清中Hcy含量均明显高于对照组。 (6);染毒30d,烟气暴露组潜伏期BDNF表达显著高于对照组(P0.05);染毒90d,中剂量烟气暴露组BDNF表达显著高于对照组(P0.05);染毒180d,烟气暴露组BDNF表达显著高于对照组(P0.05)。 短期烟气暴露可提高大鼠学习记忆功能,随染毒时间增长,学习记忆功能下降。该作用与海马BDNF的表达密切相关。
[Abstract]:To study the effects of cigarette smoke exposure on learning and memory function in rats and to explore the role of brain-derived neurotrophic factor (BDNF). Male Sprague-Dawley rats were randomly divided into control group, low dose group (10 min exposure), middle dose group (20 min exposure) and high dose group (40 min exposure). The learning and memory function of rats was measured. The indexes were as follows: body weight, peripheral blood cell parameters, serum homocysteine (Hcyl) content and HE staining to observe the pathological changes of hippocampus and the expression of BDNF in hippocampus by immunohistochemical method. Compared with the normal control group, the results were as follows: (1) the weight gain of rats decreased significantly, and the white blood cell count increased first and then decreased. The number of red blood cells decreased first and then increased, the incubation period of the smoke exposure group was significantly lower than that of the control group for 30 days, and the incubation period of the middle dose smoke exposure group was significantly lower than that of the control group for 90 days. The incubation period of the exposed group was significantly higher than that of the control group for 180 days. With the increase of time, the learning and memory function of the control group was gradually enhanced, that of the low dose group was decreased first and then increased, and that of the middle dose group was enhanced. The results of HE staining showed that there was no significant change in hippocampal area of rats exposed to cigarette smoke for 30 days or 90 days after exposure to cigarette smoke, while the content of Hcy in serum of low dose group increased from 30 days to 90 days after exposure to cigarette smoke. The contents of serum Hcy in the medium dose group and high dose group were significantly higher than those in the control group at 180 days after exposure, and the expression of BDNF in the incubation period of the smoke exposure group was significantly higher than that in the control group at 30 days after exposure. The expression of BDNF in the exposure group was significantly higher than that in the control group for 90 days, and the expression of BDNF in the smoke exposure group was significantly higher than that in the control group at 180 days. Short-term smoke exposure could improve the learning and memory function of rats, and the learning and memory function decreased with the increase of exposure time. This effect is closely related to the expression of BDNF in hippocampus.
