骨碎补总黄酮对兔激素性股骨头坏死脂代谢及病理学的影响

【摘要】:目的:本课题通过观察骨碎补总黄酮对新西兰大白兔激素性股骨头坏死血清甘油三酯、总胆固醇及各组动物脂肪细胞平均直径、骨小梁面积的影响,研究骨碎补总黄酮治疗激素性股骨头坏死的作用效果和作用机理。方法:实验采用新西兰大白兔30只(雄雌各半)作为研究对象,建立激素性股骨头坏死模型后,并于造模成功后第一天空白、模型组给予生理盐水6ml·k g-1灌胃,实验组按照35mg·kg-1给予骨碎补总黄酮溶液(6 g·L~(-1))。在灌胃8周之后,检测各组动物血清甘油三酯、总胆固醇的变化情况,解剖出股骨头并切片光镜下计算脂肪细胞平均直径、骨小梁面积,然后进行组间比较。结果:血清甘油三酯、总胆固醇的测定:空白组与模型组血清甘油三酯、总胆固醇相比,差异均有显著统计学意义(P0.05),表明通过大剂量的激素应用后模型组家兔的血清甘油三酯明显升高,总胆固醇明显升高。实验组与模型组血清甘油三酯、总胆固醇比较,差异均有显著性差异(P0.05),实验组与空白组相比,差异无统计学意义(P0.05),提示用骨碎补总黄酮干预后,血清甘油三酯、总胆固醇水平基本可以降低到正常水平。脂肪细胞平均直径的测定:模型组与空白组、实验组相比,脂肪细胞平均直径明显增大,差异均有显著性统计学意义(P0.05);实验组与空白组比较,脂肪细胞平均直径无明显变化,差异无统计学意义(P0.05)。骨小梁面积模型组与空白组、实验组比较,模型组骨小梁面积明显减少,组间差异均有统计学意义(P0.05);实验组和空白组比较,两组细胞中骨小梁面积无明显变化,组间差异无统计学意义(P0.05)。结论:骨碎补总黄酮可以调控血清甘油三酯、总胆固醇的含量,改善脂肪细胞平均直径,有改善脂肪代谢紊乱,促进堆积脂肪代谢的作用;能够刺激骨小梁的生成,最终实现防治激素性股骨头坏死的目的,为临床治疗本病提供理论依据。
[Abstract]:Objective: to observe the effects of total flavonoids of Fructus fragmentosa on serum triglyceride, total cholesterol, mean diameter of adipocytes and trabeculae area of steroid-induced femoral head necrosis in New Zealand white rabbits. Objective: to study the effect and mechanism of total flavonoids in the treatment of steroid-induced osteonecrosis of femoral head. Methods: thirty New Zealand white rabbits (male and half female) were used to establish steroid-induced femoral head necrosis model. The model group was given normal saline 6ml kg-1 intragastrically on the first day after the establishment of the model. The experimental group was given 6 g of total flavonoids solution according to 35mg kg-1. After 8 weeks of gastric perfusion, the changes of serum triglyceride and total cholesterol were measured, the femoral head was dissected and the mean diameter of adipocytes and trabecular area of bone were calculated under light microscope, and then the comparison was made between the groups. Results: determination of serum triglyceride and total cholesterol: serum triglyceride and total cholesterol were compared between blank group and model group. The results showed that the serum triglyceride and total cholesterol in the model group were significantly higher than those in the control group. There were significant differences in serum triglyceride and total cholesterol between the experimental group and the model group (P 0.05), but there was no significant difference between the experimental group and the blank group (P 0.05). Total cholesterol levels can be basically reduced to normal levels. Measurement of mean diameter of adipocytes: the mean diameters of adipocytes in model group and blank group were significantly larger than those in control group (P 0.05), but there was no significant change in average diameter of adipocytes in experimental group and blank group. The difference was not statistically significant (P 0.05). The trabecular area in the model group was significantly decreased compared with the blank group, and the difference between the two groups was statistically significant (P 0.05), but there was no significant change in the trabecular area between the experimental group and the blank group. There was no significant difference between the two groups (P 0.05). Conclusion: the total flavonoids can regulate the content of serum triglyceride and total cholesterol, improve the average diameter of adipocytes, improve the disorder of fat metabolism, promote the accumulation of fat metabolism, and stimulate the formation of trabeculae. Finally, the purpose of prevention and treatment of hormone-induced femoral head necrosis is achieved, which provides a theoretical basis for the clinical treatment of this disease.
