有氧运动对MI后大鼠心肌组织抑炎因子的影响及调控研究

【摘要】:研究目的:探讨有氧运动对心肌梗死(MI)后大鼠心肌组织抑炎因子白介素-4(IL-4)、白介素-10(IL-10)分泌情况及信号传导蛋白和转录激活物6(STAT6)磷酸化水平的影响,以及STAT6在炎症反应中的作用机制。研究方法:选用30只雄性8周龄的SD大鼠,随机分为假手术组(Sham)、心肌梗死组(MI)、梗死+运动组(ME)。以冠状动脉左前降支(LAD)结扎术构建MI模型,运动方式为间歇有氧运动。运动方案:1周的适应性运动后,10m/min热身10min,然后以20m/min运动8min,再以15 m/min运动2 min,交替4个循环,最后20 m/min运动8min,10 m/min运动2 min后结束。每天训练1次,5 d/w,共训练8 w。以二维超声心动图检测大鼠心脏室壁厚度、射血功能;采用ELISA的方法检测大鼠心肌组织IL-4、IL-10的分泌水平;Western blot检测心肌组织p-STAT6、t-STAT6的表达水平。研究结果:(1)MI手术使大鼠心脏心室壁变薄,射血功能降低,射血分数(EF)与Sham组相比明显减小(P≤0.01);8周有氧运动使MI大鼠的室壁厚度增加、射血功能加强,ME组舒张末期左室后壁厚度(LVPWd)与舒张末期室间隔厚度之和明显高于MI组(P≤0.05),ME组EF比MI组明显升高(P≤0.05)。(2)MI组心重/体重比值(HW/BW)显著低于Sham组(P≤0.05),ME组与MI组相比有明显的增加(P≤0.05);而ME组与MI组肺重/体重比值(LW/BW)相比有明显的降低(P≤0.01)。(3)ME的心肌组织IL-4分泌水平与MI组相比有明显升高(P≤0.05);ME组心肌组织的IL-10分泌水平也显著高于MI组(P≤0.05)。(4)Western blot 结果显示,ME 组的 p-STAT6/t-STAT6 比值显著高于MI 组(P≤0.05)。研究结论:(1)有氧运动有利于MI大鼠心脏舒缩功能的改善;(2)有氧运动有助于MI大鼠心功能的恢复,减轻炎症反应;(3)有氧运动可以升高MI大鼠心肌组织中抑炎因子IL-4、IL-10的分泌水平;(4)有氧运动有助于激活大鼠心肌组织STAT6的磷酸化改变,从而抑制炎症反应。
[Abstract]:Objective: to investigate the effects of aerobic exercise on the secretion of interleukin-4 (IL-4), interleukin-10 (IL-10) and phosphorylation of signal transduction protein and activator 6 (STAT6) in myocardial tissue after myocardial infarction (MI) in rats. And the mechanism of STAT 6 in inflammatory reaction. Methods: thirty male 8-week-old SD rats were randomly divided into sham-operated group, myocardial infarction group and myocardial infarction group. The MI model was established by left anterior descending branch ligation of coronary artery.
