线粒体基因复合扩增进行种属鉴定的研究

【摘要】:目的 生物性检材的种属鉴定是法医物证学主要研究内容之一。本课题针对线粒体DNA(Mitochondrial DNA,mtDNA)编码序列的保守区和可变区在人与哺乳动物间的异同,设计并建立稳定可靠的种属鉴定体系,探索一种适合法医学实践的,快速、灵敏、简便而实用的种属鉴别方法,以解决当前种属鉴定中存在的不足。 方法 针对本课题的研究目的,对mtDNA的37个基因进行研究,制定目的基因筛选原则,遵循该原则筛选出合适的编码基因,根据建立复合扩增体系的要求,,建立稳定可靠的线粒体基因复合扩增反应体系。复合扩增产物用非变性聚丙烯酰胺凝胶垂直电泳(PAGE)、硝酸银染色技术进行种属鉴定。为验证该系统的稳定性以及在法医学实践中的可行性,进行了包括系统灵敏度检测、不同环境下样本的检测、形成于不同基质上样本的检测和陈旧样本等的检测,从而对其法医学应用价值进行评估。
[Abstract]:Objective to identify the species of biologic samples is one of the main contents of forensic forensics. This paper aims at the similarities and differences of the conserved and variable regions of mitochondrial DNA(Mitochondrial mtDNAs between human and mammal. To design and establish a stable and reliable species identification system, and to explore a rapid, sensitive, simple and practical method suitable for forensic practice, in order to solve the shortcomings of current species identification. Methods for the purpose of this study, 37 genes of mtDNA were studied, and the screening principles of target genes were worked out. According to the requirements of establishing a multiplex amplification system, appropriate coding genes were screened according to the principle. A stable and reliable multiplex amplification system of mitochondrial gene was established. The products were identified by polyacrylamide gel electrophoresis and silver nitrate staining. Feasibility in the practice of forensic medicine, It includes system sensitivity detection, sample detection in different environments, sample detection on different matrix and old sample detection, so as to evaluate the value of forensic application.
