生物样品中常见毒品检测方法研究

【摘要】: 近年来,受世界毒潮泛滥的影响,我国禁毒斗争形式依然十分严重。在海洛因类毒品没有得到有效遏制的情况下,苯丙胺类毒品已经取代海洛因类毒品成为我国的主要毒品。因此,迫切需要建立一种准确、快速检测生物样品中苯丙胺类和海洛因类毒品的方法。本文分别利用微波萃取法和液液萃取法对血液和尿液中的苯丙胺类和海洛因类毒品进行了研究。 建立了人体血液中甲基苯丙胺(MA),3,4-亚甲二氧基苯丙胺(MDA),3,4-亚甲二氧基甲基苯丙胺(MDMA)的微波萃取-气相色谱(GC)和气相色谱/质谱(GC/MS)测定方法。分别考察了萃取溶剂种类、用量、pH以及萃取温度、时间等因素对萃取率的影响。实验结果表明,血液中MA,MDA,MDMA的最佳提取条件为:调节血样pH为13,以乙酸乙酯为萃取溶剂,于30℃下微波提取10 min。该方法未对药物进行衍生化,是一种快速,准确,灵敏度高的测定血液样品中MA,MDA,MDMA的方法。 建立了人体尿液中MA,MDA,MDMA的微波萃取-气相色谱(GC)和气相色谱/质谱(GC/MS)测定方法。分别考察了萃取溶剂种类、用量、pH以及萃取温度、时间等因素对萃取率的影响。实验结果表明,尿液中MA,MDA,MDMA的最佳提取条件为:调节尿样pH为12,以环己烷为萃取溶剂,于40℃下微波提取10 min。该方法未对药物进行衍生化,是一种快速,准确,灵敏度高的测定尿液样品中MA,MDA,MDMA的方法。 建立了人体血液中的可待因、吗啡、3-单乙酰吗啡(O3)的测定方法,分别考察了不同萃取溶剂、萃取时间、溶液的pH和用量对萃取结果的影响,以及不同衍生化试剂和用量对检出限的影响。该方法操作简单,大大提高了样品前处理的效率。
[Abstract]:In recent years, influenced by the flood of drugs in the world, the fight against drugs in our country is still very serious. Without the effective control of heroin drugs, amphetamines have replaced heroin drugs as the main drugs in China. Therefore, there is an urgent need to establish an accurate and rapid method for the detection of amphetamines and heroin in biological samples. The amphetamines and heroin drugs in blood and urine were studied by microwave extraction and liquid extraction respectively. A method for the determination of methamphetamine (MA), _ 3N) _ 4-methylenedioxyamphetamine (MDA), _ (3) -4-methylenedioxymethamphetamine (MDMA) in human blood by microwave extraction-gas chromatography (GC) and gas chromatography / mass spectrometry (GC/MS) was developed. The effects of solvent type, amount, pH, extraction temperature and time on the extraction rate were investigated. The results showed that the optimal extraction conditions of MA,MDA,MDMA in blood were as follows: adjusting the pH of blood sample to 13, using ethyl acetate as extraction solvent and microwave extracting 10 min. at 30 鈩,
