盆栽小菊高频再生体系建立与试管开花研究

【摘要】:本试验利用盆栽小菊花瓣作为外植体,建立以花瓣为外植体的高频快繁再生系统。试验以MS作为基本培养基,添加生长素与细胞分裂素,通过正交试验方法筛选出愈伤组织诱导、不定芽分化以及继代增殖中最优激素配方。针对不定芽玻璃化现象采用提高渗透压、减少细胞分裂素浓度、降低培养瓶内空气湿度进行调控。通过离体培养盆栽小菊花芽进行试管开花诱导,通过单因子筛选影响因素,研究不同植物生长激素在植株试管开花中的作用。研究结果表明:(1)盆栽小菊 ' Chrystal Pink'品种愈伤分化的最适培养基配方为MS+NAA1.2mg/L+6-BA1.0mg/L+TDZ 0.4mg/L,诱导率为 74.56%。'Rainbow Worth' 品种为MS+NAA1.2mg/L+6-BA1.5mg/L+TDZ0.2mg/L,诱导率为 77.21%。'Chrystal Bronze' 品种为MS+NAA0.3mg/L+6-BA1.0mg/L+TDZ0.4mg/L,诱导率为 62.23%。' Little Rock ' 为MS+NAA1.2mg/L+6-BA1.0mg/L+TDZ0.4mg/L,诱导率为 28.87%。(2)盆栽小菊品种 'Chrystal Pink'、'Rainbow Worth'、'Chrystal Bronze' 最佳不定芽诱导培养基为 MS+6-BA1.0mg/L+NAA0.1mg/L,诱导率分别为 41.11%、43.33%、51.11%。品种'Little Rock'为 MS+6-BA1.0mg/L+NAA0.3mg/L+TDZlmg/L,诱导率为 22.22%。(3)四个品种盆栽小菊 'Chrystal Pink'、'Rainbow Worth'、'Chrystal Bronze'、'Little Rock'最佳继代增殖培养基均为MS+6-BA0.6mg/L+NAA0.05mg/L+TDZ0.1mg/L。将盆栽小菊'Chrystal Pink,品种继代增殖培养基中细胞分裂素含量减半,玻璃化率降低到21.11%。综合玻璃化率与增值率两方面因素来看,蔗糖浓度提升到50g/L有利于改善玻璃苗增殖。(4)盆栽小菊品种 'Chrystal Pink'、'Rainbow Worth'、'Chrystal Bronze' 最佳生根培养基为MS+NAA0.2。'Little Rock'品种的最佳生根培养基为MS培养基。(5)蔗糖浓度为50 g/L时,开花率高,花期长。培养基中添加0.1mg/L的NAA开花率达到52.23%-65.56%。添加 0.1mg/L6-BA 开花率为 17.76%-28.89%。添加 0.5mg/LTDZ 诱导开花率为31.11%-37.78%。0.5mg/LKT对试管开花作用明显,开花率达到39.89%-43.23%。在MS培养基中添加1.0mg/LGA3时开花率达到39.89%-43.33%。
[Abstract]:In this experiment, the potted chrysanthemum flaps were used as explants to establish a high frequency rapid propagation regeneration system with petals as explants. Ms was used as the basic medium and auxin and cytokinin were added. The optimal hormone formula for callus induction, adventitious bud differentiation and subculture proliferation was obtained by orthogonal experiment. The vitrification of adventitious buds was treated by increasing the osmotic pressure and reducing the concentration of cytokinins. Through in vitro culture pot culture of chrysanthemum flower bud for tube flowering induction, through the single factor screening influencing factors, The effect of different plant growth hormone on plant in vitro flowering was studied. The results showed that the optimal medium for callus differentiation of potted chrysanthemum 'Chrystal' was MS NAA1.2mg/L 6-BA 1.0 mg / L TDZ 0.4 mg / L, and the induction rate was 74.56.Rainbow Worth' was MS NAA1.2mg/L 6-BA 1.5 mg / L. The inductive rate of MS NAA0.3mg/L 6-BA1.0 mg / L TDZ0.4 mg / L, and the inductive rate of MS NAA1.2mg/L 6-BA1.0 mg / L TDZ0.4 mg / L, 'Little Rock' was MS 6-BA1.0 mg / L TDZ0.4 mg / L, and the inducement rate was 28.877.82) the best adventitious bud induction medium was MS 6-BA1.0 mg / L NAA 0.1 mg / L, and the inducement rate was 41.11% 43.33 mg / L, respectively, in the medium of MS 6-BA1.0 mg / L NAA0.1 mg / L, and the inductive rate was 41.11 mg / L, 43.33 mg / L and 41.11% / L, respectively, and the optimal adventitious bud induction medium was MS 6-BA1.0 mg / L NAA0.1 mg / L, and the inductive rate was 28.87 mg / L, and the optimal medium for inducing adventitious buds was MS 6-BA1.0 mg / L NAA 0.1 mg / L, and the induced rate was 41.11% 43.33 mg / L, respectively. The best subculture medium for MS 6-BA 1.0 mg / L NAA0.3mg/L TDZlmg / L, inductive rate 22.222.35) is MS 6-BA 0.6 mg / L NAA0.05mg/L TDZ0.1 mg / L, and the content of cytokinin in the subculture medium is halved, and the content of cytokinin in the subculture medium is halved, and the content of cytokinin is halved in the subculture medium of Chrystal Pinkin, the best subculture medium is MS 6-BA 0.6 mg / L NAA0.05mg/L TDZ0.1 mg / L, and the content of cytokinin in the subculture medium is halved. The vitrification rate is reduced to 21.11. Considering both the vitrification rate and the value added rate, Raising sucrose concentration to 50g / L could improve the proliferation of glass-seedling. 4) the best rooting medium for MS NAA0.2.'Little Rock' was MS medium. The best rooting medium for MS NAA0.2.'Little Rock' was MS medium with 50 g / L sucrose concentration, and the flowering rate was high when sucrose concentration was 50 g / L. The best rooting medium was Chrystal Bronze' for potted chrysanthemum, and the best rooting medium for MS NAA0.2.'Little Rock' was 50 g / L of sucrose. The flowering rate of NAA with 0.1 mg / L was 52.23-65.56. The flowering rate of 0.1 mg / L 6-BA was 17.76-28.89. The induced flowering rate was 31.11-37.78.0.5mg / L LKT had obvious effect on tube flowering. The flowering rate reached 39.89- 43.23.The addition of 1.0 mg / L 6-BA in MS medium reached 39.89- 43.33.
