山西省野生花楸的园林应用价值评价与繁殖方法研究

【摘要】:本试验以山西省野生花楸为试验材料,通过对两种野生花楸的园林应用价值的评价及花楸树种子休眠原因、繁殖方法、愈伤组织诱导的研究,确定打破种子休眠、栽培繁殖的最佳方法及嫩叶快繁愈伤组织诱导的最佳激素组合。旨在为花楸树的生产提供参考,为园林景观设计提供选材,从而科学的指引花楸在山西省,甚至整个园林绿化产业的发展方向。试验得出以下结论:1.山西省野生花楸是今后园林绿化造景的重点推荐及开发利用的新品种,园林应用价值极高,目前山西省园林绿化尚未规模化应用。2.取自山西省庞泉沟自然保护区野生状态下的花楸树种子之所以休眠,主要原因是由于花楸树种子的种皮及种胚中,均含有可溶于醇类的萌发抑制物,这种抑制物浓度的增高,会使抑制种子萌发的作用增强,从而造成种子休眠。3.花楸树的繁殖以播种繁殖为主。秋季直播、赤霉素处理、低温层积处理、赤霉素处理与层积处理结合及组培室无菌培养的方法,均可以打破花楸树种子的休眠,达到发芽的目的。试验结果如下Ⅰ.不同浓度的赤霉素溶液处理花楸树的种子,其发芽率不同。经浓度为0.5 g/L的赤霉素处理过的种子发芽率最高,发芽率为22.67%。Ⅱ.由于层积地点、层积环境的可控性等条件不同,种子的发芽率也存在差异。经过2。C冰箱内层积处理的种子发茅率略高于室外低温层积处理的结果,最高发芽率可达42.80%。Ⅲ.不同的播种方法对种子发芽时间的影响不同。组培室无菌培养的花楸树种子发芽的起始时间最早,不论是新种子,还是陈种子,在播种后7d即可露白播种,减少了前期低温处理的时间,新种子的利用效率大大提高。Ⅳ.花楸树种子发芽率按从高到低的顺序排列依次为:赤霉素与层积处理结合层积处理赤霉素处理秋季直播。用0.5 g/L的赤霉素溶液处理花楸树种子,同时结合低温层积处理,可获得最佳发芽率,最高可达52.00%,平均发茅率为48.33%。4.花楸树愈伤组织诱导研究:叶片最佳HgCl2灭菌时间为6 min,花楸树嫩叶愈伤组织诱导的最佳激素组合为MS+0.1 mg/L NAA+1.0 mg/L 6-BA。

Gibberellin treatment, low temperature stratification treatment, combination of gibberellin treatment and stratification treatment and aseptic culture in tissue culture can all break the dormancy of the seeds of Liriodendron mandshurica.
