Hyal2在电击后心脏组织表达的免疫组化研究

【摘要】: 目的目前电击伤(死)案件随着电力事业蓬勃发展呈逐年增加的趋势,法医学检案中用低压电来自杀或谋杀的案件也不少见。目前国内外法医学家在生前电击伤(死)和死后电击伤的鉴别及电损伤时间的推断等方面研究取得了一定进展,但仍没有取得重大突破。为了准确鉴别生前电击和死后电击伤以及损伤时间的准确推断,我们设计了该实验。 方法1.利用220V交流电(50Hz, 150-200 mA)电击Wister大白鼠胸部(心脏体表投影位置)30秒钟复制动物模型。2.电击后在不同时间(1h、2h、3h、6h、8h、12h、1d、2d、3d、6d、12d)断颈处死,手术打开胸腔,取出心脏,PBS液冲洗后,将标本置于10%甲醛溶液中固定,作病理切片(5μm)。3.HE染色观察其组织病理学改变;用Hyal2为一抗标记心肌组织,观察其在心肌组织的分布及不同时间的变化情况。 结果: 伤后1至6小时,心肌多灶性变性坏死,变性坏死区内肌细胞广泛断裂,部分心肌细胞呈波纹状;心肌胞浆凝聚、均质变、嗜酸性变,部分心肌横纹不清,胞核固缩、深染、扭曲,部分核伸长,部分核染色质边集、核膜增厚、呈空泡状;心肌间隙增宽,水肿,部分区域散
[Abstract]:Objective at present, the electric injury (death) cases are increasing year by year with the rapid development of electric power industry, and the cases of suicide or murder with low-voltage electricity in forensic medical examination cases are not uncommon. At present, forensic scientists at home and abroad have made some progress in the identification of antemortem electric injury (death) and postmortem electric injury, and in the estimation of electric injury time, but no significant breakthrough has been made. In order to accurately identify the antemortem electric shock and postmortem electric shock injury as well as the exact estimation of injury time, we designed the experiment. Method 1. Wister rats were subjected to 220V AC 50Hz, 150-200mAshock to the chest (the projection position of the heart surface was used to reproduce the animal model for 30 seconds. After electric shock, the neck was amputated at different times (1h, 2h, 3h, 8h, 12h, 1h, 2d, 3d, 6d, 12d). After operation, the chest cavity was opened, the heart fluid was washed, the specimen was fixed in 10% formaldehyde solution, the histopathological changes were observed by pathological section with 5 渭 m).3.HE staining, and the myocardial tissue was labeled with Hyal2. Its distribution in myocardial tissue and its changes at different time were observed. Results: From 1 to 6 hours after injury, the myocytes in the zone of degeneration and necrosis were extensively broken, some of them were corrugated, the cytoplasm of the myocardium was condensed, homogenized, eosinophilic, and some of the myocardial transverse lines were unclear, and the nuclei were pyknotic and deep stained. Twisted, partially elongated, part of nuclear chromatin set, nuclear membrane thickened, vacuolated; myocardial space widened, edema, part of area dispersed.
