瘤胃丁酸灌注促进山羊瘤胃上皮乳头生长及其机理研究

  先前研究表明,瘤胃丁酸灌注可上调上皮细胞细胞周期蛋白cyclin D1,但其分子机制尚未阐明。本文的目的是研究丁酸对瘤胃上皮乳头生长的影响及其分子机制,首次探讨了瘤胃丁酸灌注对山羊瘤胃上皮细胞增殖和细胞凋亡相关基因表达的影响。研究分为两部分:1,瘤胃丁酸灌注实验;2,瘤胃上皮细胞原代培养实验。1,瘤胃丁酸灌注实验二十四只装有瘤胃瘘管的健康山羊(4月龄;体重19.05±0.43kg)随机分成两组:B组(n=12,丁酸50ml 0.3 g/kg·BW/天经瘘管灌注到瘤胃)和C组(n=12,经瘘管注入相同剂量的缓冲液,但不含丁酸)。每天上午饲喂前1h灌注,实验期为28天。每组的四只山羊分别在最后一天灌注后的5、7和9小时被屠宰。采集瘤胃上皮样品,用游标卡尺检测瘤胃上皮形态上变化;Real-time PCR法检测基因表达。采集瘤胃液,用气象色谱法测定瘤胃内挥发性脂肪酸(SCFA)含量。结果表明,与对照组相比,丁酸灌注组1小时后瘤胃内丁酸浓度增加了 99.77%,且差异显著,并且持续了2.5h。瘤胃上皮形态学数据表明,与对照组相比,丁酸灌注组山羊前腹盲囊,腹囊和后腹盲囊在5h时的瘤胃上皮的长度分别...
